As long as there are no more than five spaces between the tiles you want to match, which need to be identical, you can make that match by clicking on them, creating the line that connects them, and then having them cleared from the screen. Even more importantly, do it before the time granted for it runs out on you because if you don't match and clear all tiles before the time bar runs out, you lose and have to start again from scratch.
