In the Missions Mode, there are six missions for you to complete, each one a bigger challenge than the last. The goals remain in the same vein for all of them:

picking up a certain number of passengers and driving them to their destinations;
completing the route by picking up and dropping the customers before you run out of time to do so;
follow the arrows on the screen to drive the route according to plan;

In the first mission, you will go to six bus stops, wait for six school girls to board, and then drive them to their school before classes start!
